Here is a link to a long (two page) discussion about all the different ways to do this trip.

But quoting to make it easer...

There are two direct buses a day operated by ALSA - times and fares at www.alsa.es although be aware that on-line ticket reservation is not possible for journeys originating in Portugal.

You can travel via Faro, changing buses there and also twice a day, www.eva-bus.com

There are various other convoluted options. Please see my webpage www.algarvebus.info/001.htm for some information on train/ferry/bus options via Faro and Vila Real de Santo Antonio.

Direct train travel is only possible using the single daily train from Lisbon to Madrid (2230 arrive 0858) then the Spanish AVE high-speed train to Seville. This is also the most expensive option.

There are direct flights between Lisbon and Seville. On a good day fares are not much more than the other options. See www.flytap.com and www.vueling.com

Hope that helps.
